The custom-designed Nikon LSM A1HD25 TIRF-MFIS with Super-resolution Microscope N-STORM is mounted on an inverted Nikon Ti2-E microscope. The microscope is equipped with the PicoQuant FLIM system "HydraHarp400", and a Multiparameter Fluorescence Imaging Spectroscopy (MFIS) extension with six detection channels developed in the Seidel group.

Available laser lines for continuous wave (cw) excitation from LU-N4 LASER UNIT are: 405 nm, 488 nm, 561 nm and 640 nm.

Available laser lines for pulsed and cw excitation, e.g. for FLIM experiments are: 485 nm, 640 nm and White Light Laser NKT SuperK Varia.

In addition to the confocal detection unit, this system also contains a TIRF-module, which can be used with the internal lasers. For detection in TIRF-mode, a Hamamatsu Orca Fusion-BT three cameras are used.

The system contains an on-stage Okolab Incubator H201-ENCLOSURE-BLA with Active Humidity Controller.

Possible applications for this system are: confocal (live cell) imaging, including z-stacks, movies and time-lapse experiments, photobleaching approaches such as Fluorescence Recovery After Photobleaching (FRAP) and related techniques and also Fluorescence Correlation Spectroscopy (FCS). The system is well suited for fluorescence lifetime measurements, e.g. in FRET applications and, as a unique feature, for multiparameter fluorescence detection in confocal images.
